Definition

Das HNF1B-Gen ist auf Chromosom 17q12 lokalisiert und kartiert für den „Hepatocyte nuclear factor-1-beta“ auch als „Transcription factor-2“ bezeichnet, ein Transkriptionsfaktor der bereits beim Föten in Nieren Leber, Gallengängen, Thymus, Genitaltrakt, Pankreas, Lunge und Darm exprimiert wird.

Die autosomal dominante monogene Mutation des HNF1B-Gens führt zu MODY5 ("Maturity-onset Diabetes of the Young type 5"), einem Krankheitsbild aus der Gruppe autosomal-dominant vererbter, klinisch heterogener, nicht immer insulinabhängiger Formen des Diabetes.

Mehrere krankheitsverursachende Keimbahnmutationen sind für folgende Erkrankungen nachgewiesen:

  • Diabetes mellitus, noninsulin-dependent OMIM: 125853 (Furuta H et al.)
  • Renal cysts and diabetes syndrome (Nierenzysten und Diabetes-Syndrom) OMIM: 137920 (Bingham C et al. 2003)  
  • Renal cell carcinoma, chromophobe OMIM: 144700 (Rebouissou et al. 2005)
  • CAKUT: Wakayama et al. (2010) identifizierten heterozygote HNF1B Mutations in 5 (10%) von 50 japanischen Kindern mit kongenitalen Anomalien von Nieren und ableitenden Harnwegen (CAKUT), die teilweise mit Nierenzysten kombiniert waren.
