Definition

Als Interleukine (von lat/griech. inter = zwischen; leukos = weiß; kinein = sich bewegen) wird eine Gruppe von körpereigenen, kurzkettigen Peptidhormonen (Zytokine) des Immunsystems bezeichnet. Interleukin-3 ist ein Glykoprotein das v.a. von Antigen- und Mitogen-aktivierten T-Lymphozyten gebildet wird. IL-3 hat einen starken Einfluss auf Wachstum und Differenzierung hämatopoetischer Stammzellen. Es bestehen synergistische Effekte zu Interleukin-4 und GMCS.

Allgemeine Information

Die Wirkung des Interleukins-3 wird über Interleukin-3-Rezeptoren vermittelt. Diese werden auf Makrophagen, Mastzellen, Endothelien, hämatopetische Vorläuferzellen im Knochenmark, neutrophilen Granulozyten sowie verschiedenen Leukämie-Zellen exprimiert. Die Rezeptordichte liegt bei 1000–5000/Zelle. Sie reduziert sich mit zunehmender Reifung der Zellen.

Die Signaltransduktion beruht auf der Phosphorylierung des Rezeptors und der konsekutiven Aktivierung der Proteinkinase C.

 

Therapie allgemein

Therapeutisch wird Interleukin-3 zur Stimulation der Blutbildung nach einer Chemotherapie eingesetzt. Einsatz auch nach Knochenmarks- oder Stammzelltransplantation. 

Hinweis(e)

Polymorphismen im Interleukin-3-Gen führten in einer chinesischen Population zur Schizophrenie. Cyclosporin A und Glukokortikoide hemmen die Produktion von IL-3.
